Multi-Dot Enzyme-Linked Immunosorbent Assay for Serodiagnosis of Helminths


Abstract
In this study, it is attempted to test the reliability and simplicity of multi-dot ELISA method utilizing a nitrocellulose membrane strip dotted with various helminths crude antigens.
Antibodies in helminthic patient sera were obtained from 7 cases such as clonorchiasis, schistosomiasis, paragonimiasis, fascioliasis, plerocercoidosis 1, 2 and 3.
The reflectance of the spots of antigen-antibody reaction were measured with a dual-wave length TLC chromatograph scanner(560nm).
Antibodies in helminthic patient sera were clearly detected on the strip as bluish purple spots.
Densities of spots were appeared the highest in clonorchiasis with 1,474.000, schistosomiasis with 102.000, paragonimiasis with 1,774.000 fascioliasis with 993.000, plerocercoidosis with 1.785.750, 2.267.750 and 3.880.250.
The advantages obtained in process and method of this experiment here was suggest that crude antigen can be used without losing reliability, inspite of small quantities of antigens used (1§¡ of mg/§¢ antigen solution per dot), the antigen dotted
nitrocellulose membranes were using to store and stable at least for 6 months at 4¡É and the results can be obtained within 3 hours.
